Scientists Uncover Key Step in How Diazotrophs “Fix” Nitrogen

Nitrogen is an essential component in the production of amino acids and nucleic acids — both necessary for cell growth and function. Although the atmosphere is composed of nearly 80% nitrogen, this nitrogen is in the form of dinitrogen (N2), which cannot be processed by most organisms. Atmospheric nitrogen must first be converted, or “fixed,” into a form that can be used by plants, often as ammonia.

There are only two ways of fixing nitrogen, one industrial and one biological. To better understand a key component of the biological process, University of California San Diego Professor of Chemistry and Biochemistry Akif Tezcan and Assistant Professor of Chemistry and Biochemistry Mark Herzik took a multi-pronged approach. Their work appears in Nature.

Thriving in a hostile environment

The industrial method is through the Haber-Bosch process, the advent of which in the early 20th century enabled the large-scale production of synthetic fertilizers, increased agricultural yields and helped the world population skyrocket. However, this process is energy-intensive, operating at very high temperatures and pressures. It also requires vast amounts of hydrogen, generated by burning fossil fuels, and is responsible for immense amounts of greenhouse gas emissions.

The biological route to fixing nitrogen is done by diazotrophs, nitrogen-fixing bacteria that contain an enzyme called nitrogenase. In contrast to the Haber-Bosch process, nitrogenase can be catalyzed at ambient pressures and temperatures and does not cause greenhouse gas emissions.

However, nitrogenase also requires large amounts of biochemical fuel, in the form of energy-producing adenosine triphosphate (ATP). Most diazotrophs make large amounts of ATP through cellular respiration by “burning” oxygen — even though nitrogenase is extremely sensitive to oxygen. Scientists have long wondered how a diazotroph, which needs to produce ATP by burning oxygen, also protects nitrogenase from being damaged by oxygen.

Diazotrophs have safety mechanisms in place; however, even with these protections in place, some oxygen penetration into the nitrogen-fixing cell is unavoidable. When this occurs, some diazotrophs employ a “method of last resort” to protect nitrogenase from destruction, called the conformational protection mechanism.

Here an iron-sulfur protein named FeSII senses increasing amounts of oxygen in the cell and binds to the nitrogenase enzyme complex, protecting it from damage and halting ammonia production. When the oxygen levels in the cell decrease, FeSII senses this again, disengages from the nitrogenase, and the ammonia production resumes.

Although scientists have long-known of the FeSII protein, the mechanism by which it protected nitrogenase from oxygen damage was not understood. To answer these questions, the researchers employed several techniques, which together provided detailed insight into how this mechanism worked.

From pineapples to AI: BT Young Scientist projects wow on first day of competition

Opening the 60th BT Young Scientist and Technology Exhibition at the RDS, President Michael D Higgins emphasised the importance of using science for good and not for the development of technology such as weapons
Ryan Foley (14), from De la Salle College in Dublin, explained that his project “Dyslexia Learning AI” uses artificial intelligence to support classroom learning and aids students with preparation for exams.The second-year student said: “I wanted to make an AI app that could help kids with dyslexia by utilising their learning styles. People with dyslexia learn in different ways. So, for example, different colours can impact how they read things, or the use of text-to-speech can help them understand concepts better.”Several of Ryan’s friends have already used the program to study for in-class exams. It allows users to play around with the background colour of inputted text to help find which one works the best for them.A text-to-speech facility uses human voices to read out text inputted into the program. The most popular features are the “rephrase” and “simplify” options to alter text.Ryan’s maths teacher Ms Kelly said these facilities could be of great support to the dyslexic students she teaches.“It can break large portions of text to help them better understand questions making learning much more accessible,” she said.Ryan was quick to emphasise that he did not want the program to simply do the work for students.“I wanted to create something that helps you understand the problem rather than just solving it. It is a completely under-utilised resource and I wanted to see if I could make a change through this project.”The transition-year student researched the increase in biodiesel production using various crops in Brazil before experimenting with seaweed.“Soya beans and other crops need arable land or freshwater to grow, in comparison to seaweed – it grows in the ocean and is easy to harvest,” Klara said.The student used steam and hexane distillation to extract oils from the seaweed. To produce biofuel, she added methanol and a catalyst to the oil extracted from the seaweed.“Donegal is a key area due to its extensive coastline and favourable conditions for seaweed growth,” she said.“It hosts 60pc of Ireland’s seaweed growth in comparison to 10pc of growth in Kerry and 20pc in Galway.”Michaela Gillooly’s project, Own your Cycle, Own Your Game, investigated the negative stigma around females speaking about their periods and the impact their menstrual cycle has on their athletic performance. The 18-year-old student got the inspiration for the project because she is a rugby player with Craig RFC in Galway.While tracking her period on the Flo app, she noticed that on the days she had her period she underperformed during a match or training.Michaela is also an under-age rugby coach and saw first-hand how embarrassed the girls she coaches were when talking about their periods.“They would approach me and tell me they were feeling unwell and would ask could I let other coaches know rather than them letting the coaches know themselves,” Michaela said.For her study, she organised two sets of fitness tests. Her first test was on female students in her school and her conclusion was periods were having a negative impact on their performance. Her second test was on a ladies’ Gaelic football (LGFA) team, which returned similar results.“I sent the results back to the LGFA team and it was clear some of the players weren’t as strong on their periods,” she said.“They could then make dietary changes such as including more calcium and iron in their diet before a match.”The initial project won the best project award at the SciFest regional competition in TUS Athlone.For her project, Starry Nights or Missed Delights?, Michelle Hughes (17) of Our Lady’s Bower, Athlone, produced two illustrated books targeting young children to foster an interest in astronomy. “There is a lack of knowledge and interest in viewing astronomical events. Targeting younger kids can help this,” Michelle said.Cathal Fitzgerald (16) and Donnacha Hartnett (15), from Mercy Secondary School Mounthawk in Tralee, Co Kerry, investigated how to use “Pineapple Power” to assist in accelerated burn recovery.“We saw online that the properties of pineapples had great potential and it went from there,” Cathal said.“We are hoping to work on this project into the future and see where it goes.”Opening the exhibition at the RDS yesterday, President Michael D Higgins emphasised the importance of using science for good and not for the development of weapons.“It is both an exciting and a morally challenging time to be a young scientist. Macros seem to come up often in the corners of the internet and social media devoted to people trying to lose weight, improve their health, look better, and feel better about themselves. But what the heck is a macro?

With more information than ever available at your fingertips, and more diets out there than you have fingers and toes to count on, it’s no wonder you might be confused. As an exercise science specialist interested in physical health and nutrition, I’ve got you covered.

“Macros” is just a shorthand term for macronutrients: protein, carbohydrates, and fats. They’re the nutritional building blocks that all foods are made up of in various ratios.

If you look at a nutrition label, you’ll see that macros are measured in grams. But it’s important to understand that the calories per gram for each macro aren’t the same. Protein and carbohydrates each have 4 calories per gram, while fat has 9 calories per gram. In other words, fat provides more than twice the amount of energy per gram compared with protein and carbohydrates.

People rarely eat proteins, carbohydrates, or fats in isolation. For example, while chicken is widely considered a source of protein, it also contains fat. Almost every food contains more than one macronutrient.

What macros do and where to find them

Beyond its job building muscle, protein also plays other critical roles in the body: as a component of enzymes, transporting nutrients, and producing hormones. Sources of protein include animal meats, eggs, fish and seafood, and dairy. While animal sources have the highest protein content, plant food sources, such as whole grains, legumes such as beans, and nuts, and seeds also contain protein. You don’t need to consume animal products to get adequate protein in your diet.

The 2020-2025 federal dietary guidelines for Americans recommend at least 46 grams of protein for adult females and at least 56 grams for adult males, although this may be too low for older adults, pregnant and lactating women, and people with high levels of physical activity.

Carbohydrates are the body’s preferred energy source. They’re found in bread, rice, pasta, fruits, dairy products, legumes and starchy vegetables. Simple sugars also fit into the carbohydrate category, and those are the ones to limit.

As for fats, there are different types: polyunsaturated, monounsaturated, saturated, and trans fat. Polyunsaturated and monounsaturated have the greatest health benefits and are found in things such as nuts and seeds and fish. Omega-3s are essential fatty acids that can reduce the risk of cardiovascular disease and are found in fatty fish such as salmon and nuts such as walnuts.

Artificial trans fats are created by an industrial process. You’ll see them listed as partially hydrogenated oils on nutrition labels for fried and baked goods. They increase the risk of heart disease and death.

Do calories matter?

In a word, yes, calories matter. But calories from different sources do affect the body differently.

For example, 2,000 calories of junk food and soda are not going to help with appetite control the same way that 2,000 calories of fruits and vegetables, whole grains, healthy fats and lean proteins will. Fiber, for example, is found in fruits, vegetables and whole grains and has been found to be a potent appetite regulator.

The good news is that the most important thing for weight management is caloric balance and consistency. In fact, evidence suggests that there is no “optimal” diet that can be applied to everyone, and that various weight loss strategies – different macronutrient distributions, intermittent fasting and so on – all balance out over the long run.

REI is still finalizing its financial results for 2024, but Artz said there are signs of improvement since last year’s results. For example, the company is nearly breaking even for both pre-dividend operating income and free cash flow, he said.
“At the same time, we still have more work to do to return the co-op to sustainable, profitable growth, which brings me back to today’s decision,” Artz wrote.

I have an important update to share today. Earlier this morning we shared with our Experiences team that after a thorough review and careful consideration, I have made the difficult decision to exit the Experiences business altogether, effective this week.
The reality is a thriving co-op requires a sustainable economic model that is capable of investing at the appropriate level to fully fund our most critical strategic ambitions. While we are still in the process of finalizing 2024 results, our preliminary financials indicate we will be close to breakeven for both Pre-Dividend Operating Income (PDOI) and Free Cash Flow (FCF).
This is a significant improvement over 2023, driven by our intentional focus on increasing full-price sales mix and gross margin, effectively managing inventory to improve in-stocks and inventory turns and carefully managing costs to optimize our spending. I greatly appreciate the work by teams across the co-op to deliver these improvements in 2024, which is a positive step in our journey to return to a healthy economic model. At the same time, we still have more work to do to return the co-op to sustainable, profitable growth, which brings me back to today’s decision.
First, I want to acknowledge and extend my gratitude to the Experiences team for all they have done and accomplished over the 40 years of its existence. You have done what has been asked of you, you have worked extremely hard, you have brilliantly educated and created wonderful experiences for nearly one million people over those years. Perhaps most importantly, I want you to know from me that you have done nothing wrong when it comes to the choice I am sharing today.
Second, it is important we all understand the economics of the Experiences business. Experiences served 40,000 customers in 2024—less than 0.4% of all co-op customers—and costs significantly more to run than it brings in. When we look at the all-up costs of running this business, including costs like marketing and technology, we are losing millions of dollars every year and subsidizing Experiences with profits from other parts of the business. Even at our peak in 2019—our best year for Experiences ever—we did not generate a profit.
Third, we have gone through many iterations and explored multiple options to keep this business up and running and to preserve jobs. We’ve held out as long as possible, but the fact remains that Experiences is an unprofitable business for the co-op, and we must adjust course. Every path to profitability we explored would have required us to invest more time, effort and focus away from parts of the business that reach significantly more customers, drive more positive financial outcomes, and have greater impact on our mission to get people outside.
Commitment to Our People
While this is the right decision for the future of our business, it will have a significant impact on people. Specifically, the 180 full-time employees and 248 part-time guides in our Experiences division whose jobs will be eliminated as we close this business.
We have some of the best Experiences guides and staff in the business and are committed to making this transition as smooth as possible.

Every full-time employee whose job is eliminated will continue to receive their regular salary through March 9, 2025, and remain active on benefits through the end of March. They will also be eligible to receive separation benefits including severance, healthcare coverage continuation via COBRA, and outplacement service support.
Shared employees (employees who split time between Experiences and Stores) will have a conversation with their store manager later today to learn what options exist for each employee to continue employment in a store capacity based on their employment status and ability to meet the necessary availability standards.
Part-time employees will remain benefits-eligible through January 2025 and will also be eligible for a severance payment.
All impacted employees will receive a Summit payout for 2024, depending on actual co-op performance vs. plan and remain eligible for future roles at the co-op.

Commitment to our Customers and Partners
This week we will also begin updating customers and partners. Customers currently booked on trips and day programs will receive a full refund of all costs paid to REI and we will work to address any associated non-refundable expenses as appropriate.
We know this decision will also significantly impact many of the travel partners that we work with across the U.S. We will begin informing partners this week and will work with them to terminate our existing contracts.
Focusing on the Core
Given the current business landscape and our strategic priorities, we must invest selectively, focusing our efforts in the areas that align most closely with our long-term commercial goals and set us up to deliver on our mission and purpose for another 86 years.
We continue to believe there is a role for REI in outdoor education and expertise. However, we need to reexamine and rework how we deliver classes and education to ensure they’re relevant to our customers, aligned to our mission and financially viable. We are funding a small team to innovate and test in this area in 2025. We also continue to believe in the importance of local community-building. We will continue to invest in local marketing and customer engagement, and the local marketing team will move back into the marketing organization to unify similar bodies of customer-facing work.
Our goal must always be the long-term financial health of the co-op, and while today’s decision is a difficult one, it is necessary. Reinvesting our resources into these core areas will enable us to drive healthy, profitable growth for the co-op in the future.
Specifically, we will be focusing our resources in areas such as:

Driving growth in our priority activities (Camp and Backpack, Run, and Hike and Outside Life) in service of our core customers and best members.
Investing in tools to help us more effectively manage inventory, which will improve sales, profitability and liquidity by driving improvements in in-stocks, locally relevant assortments and inventory flow.
Continuing to enhance the customer experience online and in stores with investments in areas including personalization and visual merchandising.

Our roots are in the gear and apparel we sell and the outdoor moments they enable. This has been the core of our business for 86 years—and I believe when we stay focused on what we do best, we can and will succeed.
As a cooperative, we are organized to drive a healthy, profitable business that allows us to serve our members, our employees and society, and fully fund commitments like the annual Member Reward, employee profit-sharing and support for nonprofits. We’ll share more about our long-term plan to do so next month.
I know that this is difficult news. The next few weeks will continue to be tough as we unwind an iconic part of our business and say goodbye to colleagues. Teams whose work is impacted should work with their leaders to understand what comes next. Today should be about processing this news and caring for impacted colleagues.
